Kisspeptin

Hospital research only; repeated dosing desensitizes the hormone system.

The signal the brain uses to start releasing reproductive hormones. A research tool with promising hospital studies, mostly from one London group. Also called Kisspeptin-10, Kisspeptin-54 and Metastin.

What the evidence shows

Triggering egg maturation in IVF
Early
In a trial of 62 women at high risk of ovarian overstimulation, mature eggs were collected from 98%. It was not compared with the standard trigger.
Low desire in men
Early
In a lab study of 32 men, an infusion increased brain and penile responses to sexual images. It measured no real-world outcomes.
Restoring lost periods
Early
In a small study of women whose periods had stopped from stress or low weight, twice-weekly doses still raised reproductive hormones after eight weeks, but periods did not return.
Raising testosterone
Early
A single dose raises the signal hormone LH for a few hours.

Side effects and risks

Unknown
Repeated or continuous dosing desensitizes the hormone system it is meant to stimulate.
Unknown
FDA says safety data for kisspeptin-10 are lacking.

Who should avoid it: Use outside a clinical study.

Athletes: Prohibited at all times (class S2). Added in 2024 as an example of testosterone-stimulating peptides, which are prohibited in males. USADA advisory on 2024 WADA list, 2023-10

How it works

Where it acts
Kisspeptin receptors on the brain cells that release GnRH.
What it changes
It triggers GnRH, which releases LH and FSH, the hormones that drive the ovaries and testes.
What people notice
In studies, an LH surge that can trigger egg maturation in IVF. Effects on desire have been studied in small groups.

How it has been given in studies

By infusion or injection in a hospital research unit. Kisspeptin-10 has a half-life of about 4 minutes and kisspeptin-54 about 28. There is no consumer dose.

Calls the craze unfounded. He found no randomized human trial showing the advertised benefits for any of the unapproved peptides he reviewed, and warns about stacking several at once. He points to animal work in which TB-500 sped up dormant tumor growth and to a theoretical cancer concern with growth-hormone peptides and BPC-157. He treats the GLP-1 drugs as the well-studied exception.

Devoted a full episode in 2024 to how these peptides are thought to work and how people use them. He presents possible benefits for healing, growth hormone, sleep and libido, and also says most of the data comes from animals, that black-market products may be contaminated, and that anyone with a tumor or cancer concern should avoid peptides that promote growth or new blood vessels.

Kisspeptin: the master regulator of reproduction?

Alexander Comninos and Waljit Dhillo

Researchers trace kisspeptin from a melanoma gene to the top of the reproductive hormone chain, and discuss possible therapies for fertility disorders.

Egg maturation trigger in IVF

Who
Women having IVF, studied as an alternative to the standard hCG trigger because of concern about ovarian hyperstimulation.
Evidence
One Imperial trial of 53 women in 2014. Eggs matured at every dose and ten women had healthy babies. No head-to-head comparison in that study.
Route
Clinical trials at Imperial College London and Hammersmith Hospital

Low sexual desire in men and women

Who
Adults distressed by low desire. Options are few, especially for men, who have no licensed drug.
Evidence
Two small crossover trials of 32 people each using one hospital infusion and brain scans. No long-term or at-home treatment data.
Route
Research trials only

Hypothalamic amenorrhea and polycystic ovary syndrome

Who
Women whose periods have stopped or who do not ovulate, where restoring natural hormone pulses is the aim.
Evidence
Hormone-response study with six women in each group using a longer-acting agonist. In women with hypothalamic amenorrhea the hormone surge came earlier than in healthy women. Preliminary: no ovulation or pregnancy outcomes.
Route
Research trials only

Legal last-resort routes

Expanded access (compassionate use)

For a serious or life-threatening condition with no comparable alternative and no trial you can join. Your physician asks the company for the drug, then files with FDA and an ethics board. The company can refuse. Costs are often not insured.

Source: FDA, expanded access for patients

Right to Try

For a life-threatening condition after approved options are exhausted and no trial is open to you. The drug must have finished Phase 1 and be in active development. Your doctor asks the manufacturer directly. FDA does not review requests.

Trial finders

Search ClinicalTrials.gov by condition or drug name, check eligibility and call the listed site. ResearchMatch is a free NIH-funded registry that lets researchers find you. Discuss risks with your own clinician before enrolling.

Personal import for serious conditions

FDA may not object when a US citizen imports up to about three months of a drug for a serious condition with no effective US treatment, with written personal-use confirmation and a US doctor's details. It is discretion, not a right.

Patient-specific compounding

When the approved product cannot meet a specific need of an identified patient, a prescriber can order a compounded version from a licensed pharmacy. Lower price alone does not qualify. Ingredients must be pharmaceutical grade.
