Matrixyl and Argireline

Subtle effects at best, mostly in company-funded studies.

The two best-known peptides in skin creams. Matrixyl is meant to signal skin to make collagen, and Argireline is sold as a mild muscle relaxer for expression lines. As cosmetics, neither had to prove it works before sale. Also called Palmitoyl pentapeptide-4, Acetyl hexapeptide-8 and Signal peptides.

What the evidence shows

Fine lines and wrinkles
Early
Small trials, many unpublished or run by ingredient makers. In one, a 10% Argireline cream cut wrinkle depth by about 30% in 10 women over 30 days. A study by the Matrixyl maker reported 18% shallower folds after 28 days.

Side effects and risks

Common
Occasional irritation. Published safety data are thin.

Who should avoid it: Nobody in particular. Patch-test if your skin is reactive.

How it works

Where it acts
Matrixyl signals skin cells to make collagen. Argireline interferes with the nerve signal to small face muscles.
What it changes
Small changes in skin collagen and in the depth of expression lines, mostly in company-funded studies.
What people notice
Subtle at best, and only where the cream is applied.

How it is used

Applied to the skin once or twice a day. Trials ran from four weeks to four months.

Paraphrased from the linked sources.

Confirmed Argireline acts on the same protein as botulinum toxin but doubted a serum penetrates deep enough for visible or lasting wrinkle results. Suggested it might help excess sweating instead.

Described Argireline as a reasonable topical add-on or non-invasive option, but far weaker than injected neurotoxins because the large molecule struggles to penetrate skin. Rated risks as minimal.

Legal last-resort routes

Expanded access (compassionate use)

For a serious or life-threatening condition with no comparable alternative and no trial you can join. Your physician asks the company for the drug, then files with FDA and an ethics board. The company can refuse. Costs are often not insured.

Right to Try

For a life-threatening condition after approved options are exhausted and no trial is open to you. The drug must have finished Phase 1 and be in active development. Your doctor asks the manufacturer directly. FDA does not review requests.

Trial finders

Search ClinicalTrials.gov by condition or drug name, check eligibility and call the listed site. ResearchMatch is a free NIH-funded registry that lets researchers find you. Discuss risks with your own clinician before enrolling.

Personal import for serious conditions

FDA may not object when a US citizen imports up to about three months of a drug for a serious condition with no effective US treatment, with written personal-use confirmation and a US doctor's details. It is discretion, not a right.

Patient-specific compounding

When the approved product cannot meet a specific need of an identified patient, a prescriber can order a compounded version from a licensed pharmacy. Lower price alone does not qualify. Ingredients must be pharmaceutical grade.

Is Argireline a muscle relaxer?

It is sold as a mild one: it interferes with the nerve signal to small face muscles. In one small study, an Argireline cream cut wrinkle depth by about 30% in 10 women over 30 days.

What does Matrixyl do?

Matrixyl is meant to signal skin cells to make collagen. A study by the Matrixyl maker reported 18% shallower folds after 28 days.
